Description
CRISPR-Cas9: programmable gene-editing system. sgRNA (single-guide RNA) directs Cas9 nuclease to target DNA via Watson–Crick base pairing. PAM (protospacer-adjacent motif, e.g. NGG) is required for recognition. Cas9 makes blunt double-strand breaks 3 bp upstream of PAM. Cellular repair (NHEJ or HDR) enables knockouts or precise edits.
