For stubborn stains I will open my bathroom window and shut the door and leave it to set in for 20-45 minutes. Separately (don't mix) you may want to try starting by using white vinegar to actually kill the mold as I believe bleach only is useful for surface level mold. This is not so much to kill the mold as it is to clean the tile so the new caulk will stick.
