I think it should work only because I regularly mirror my phone to my TV and I can display both portrait and landscape. Of course I don't turn my TV on end but I'd be surprised if there was a sensor in the TV to detect that anyway. Based on the image you sent, it looks like you're holding your phone in the normal portrait orientation.

If I play a video from my phone while mirroring, it will auto-rotate into landscape orientation and fill the TV screen, as you'd want it to. However, if I use the camera to shoot video while screen mirroring, it's always shown on the TV in portrait orientation, even if the phone is rotated so that it should be landscape. I can use Facetime in landscape mode on my iPhone, but landscape mode doesn't work in Facetime when I screen mirror either via Airplay or a Lightning.

It wouldn't be a Windows thing, but dependent on your software, if at all. Like an image editor could, in theory, put a full size view on monitor one, and then you could be zooming in and editing on monitor two. I don't know that any of them do support that, but it's plausible.
